Title: DVAX
Post by: [D] Investor on August 24, 2006, 09:49:28 AM
Judging strictly from the chart, DVAX is breaking out of its downtrend and on the verge of an upswing. DVAX's medium and long term charts show cyclic upswings and downswings like a roller coaster. Now seems to be the time for another ride UP! ;)

Waiting for a volume surge...

GREAT DAY TODAY ! SOME NEWS........

NEW ENGLAND JOURNAL OF MEDICINE Reports Positive Results From Dynavax' Ragweed Allergy Therapy Trial
Wednesday October 4, 5:00 pm ET

BERKELEY, Calif., Oct. 4 /PRNewswire-FirstCall/ -- The NEW ENGLAND JOURNAL OF MEDICINE (Vol. 355, October 5, 2006, No.14), today reported that a new approach to allergy therapy not only reduced the acute allergic responses of individuals with ragweed allergies but also sustained that effect for over 12 months. The novel treatment, called "AIC" in the paper, is a TLR9 agonist linked to ragweed allergen, developed by Dynavax Technologies Corporation (Nasdaq: DVAX - News).

Dr. Peter Creticos, principal investigator and lead author of the paper, entitled, "Immunotherapy with a Ragweed-Toll-like Receptor 9 Agonist Vaccine for Allergic Rhinitis," said that the pilot study "appears to offer a means of achieving long-term clinical efficacy in ragweed allergic rhinitis as the clinical effects suggest the induction of long-lasting disease modification. Furthermore, the demonstrated therapeutic properties and safety pave the way for a therapeutic intervention that is qualitatively superior to standard immunotherapy." Dr. Creticos is Associate Professor of Medicine and Clinical Director of the Division of Clinical Immunology of The Johns Hopkins University School of Medicine. He serves as Medical Director of the Johns Hopkins Asthma and Allergy Center in Baltimore, Maryland.

In the paper, Dr. Creticos pointed to statistically significant efficacy results including peak Nasal Symptom Complex Score (NSCS) reductions in AIC-treated patients of 55% (p=0.03) in 2001 which persisted through the 2002 ragweed season (53% reduction in NSCS, p=0.02) with no additional therapy. Additionally, the AIC-treated group used no relief medication at all during the second season, while placebo patients used antihistamines for 8 days (average) and decongestants for 4 days (average) of the two-week peak season. The intervention also generated clinically significant quality of life improvements for patients. Dr. Creticos added that the intervention was safely tolerated as no treatment-associated serious adverse reactions were reported, nor did any lab tests show abnormalities in the patients tested.

With funding from the National Institute of Allergy and Infectious Diseases and the Immune Tolerance Network, and the study drug provided by Dynavax, Dr. Creticos conducted a blinded, randomized, placebo-controlled, clinical trial in 25 ragweed allergic patients beginning in May 2001 and concluding in October, 2002. Patients were treated with either the drug or placebo, using a 6-injection regimen, prior to the first ragweed season, and were followed for two years. Dynavax supplied the study drug, now known as TOLAMBA(TM), and contributed to trial design, but did not participate in data accrual, analysis, or funding of the trial. TOLAMBA, consisting of a TLR9 agonist linked to a specific ragweed allergen, is currently being evaluated in late-stage clinical trials for the treatment of allergic rhinitis.

DYNAVAX Clinical Trials Update

In the paper, Dr. Creticos recommends additional large-scale studies, which are now underway at Dynavax. Since the reported study's initiation in 2001, Dynavax has generated a substantial amount of data in 14 clinical trials in the U.S., France, and Canada. More than 7,000 TOLAMBA injections have been administered to over 1,100 patients. In these trials, TOLAMBA was shown to be safe and well tolerated, to provide measurable improvements in allergy symptoms, and to reduce medication use.

TOLAMBA consists of 1018 ISS, a TLR9 agonist, linked to the purified major allergen of ragweed, called Amb a 1. The linking of ISS to Amb a 1 ensures that both ISS and ragweed allergen are presented simultaneously to the same immune cells, producing a highly specific and potent effect suppressing the Th2 cells responsible for inflammation associated with ragweed allergy. Moreover, this treatment reprograms the immune response away from the Th2 response and toward a Th1 memory response so that, upon subsequent natural exposure to the ragweed allergen, long-term protection is achieved.

Other Clinical Results; Trial Background

A Dynavax-funded, 30-center, placebo-controlled study in 738 ragweed allergic subjects, aged 18 to 55 years, is expected to produce interim data at one-year in the first quarter of 2007. The study known as "DARTT" (Dynavax Allergic Rhinitis TOLAMBA Trial) randomized subjects into three arms: the same dosing regimen that was used in the completed Phase 2/3 trial; a higher total dose regimen; and placebo. Subjects received six doses over six weeks prior to the start of the 2006 ragweed season. Ragweed symptoms were followed over the 2006 ragweed season and will also be followed through the 2007 season. The primary endpoint is reduction in total nasal symptom scores (TNSS) during the second (2007) peak ragweed season. Dynavax anticipates that data from the DARTT interim analysis, if positive, combined with the safety and efficacy data from the recently completed two-year Phase 2/3 trial, and from an ongoing trial in ragweed allergic children, could provide sufficient patient data for determining the potential timeline to registration for the intervention.

Additionally, Dynavax is evaluating TOLAMBA in a three-year, 19-center, pediatric trial with over 300 patients, ages six to 15 years. The primary endpoint of the study is reduction in TNSS during the 2006 peak ragweed season; a key secondary endpoint is the prevention of progression to asthma. The study was initiated in early 2005. Primary endpoint data for the study is expected in early 2007.

In January 2006, Dynavax announced that results from a two-year Phase 2/3 clinical trial of TOLAMBA showed that patients treated with TOLAMBA experienced a statistically significant 28.5% reduction in total nasal symptom scores (TNSS) compared to placebo-treated patients in the second year of the trial (p=0.024). Results also showed significant clinical benefit relative to secondary endpoints, including composite hay fever symptoms and ocular effects, and a significant reduction in antihistamine use (p=0.01). These results were achieved after a single short course of therapy prior to the first ragweed season (2004), and demonstrated that a booster dose prior to the second season (2005) was not required to achieve clinical benefit. The safety profile of TOLAMBA was favorable; systemic side effects were indistinguishable from placebo and local injection site tenderness was minor and transient.

TOLAMBA represents the foundation of a comprehensive allergy franchise for Dynavax, and has the potential to be a novel entrant in the multibillion- dollar global allergy market. In the U.S. alone, approximately 40 million people suffer from allergic rhinitis. Ragweed is the single most common seasonal allergen, affecting up to 75% of those with allergic rhinitis, or 30 million Americans. Current therapeutic options are mainly limited to symptomatic therapies and conventional allergy immunotherapy, which generally requires 60-90 shots over three to five years and represents a significant treatment burden for allergy sufferers. Dynavax believes that TOLAMBA has the potential to become the first of several new and important disease-modifying therapeutic options for patients and physicians.

Questions Arise After Dynavax Offering

By Marc Lichtenfeld

Click here for more stories by Marc Lichtenfeld

Some investors in Dynavax (DVAX - commentary - Cramer's Take) must be marveling at their good fortune, at least those who were able to buy shares of the Berkeley, Calif.-based biotech company at $4.40 in a hastily arranged stock offering on Wednesday, Oct. 4.

Imagine their glee when the stock opened Thursday at $6.35 and closed at $7.70 after a New England Journal of Medicine article was released Wednesday night stating that Dynavax's allergy therapy "appeared to offer long-term clinical efficacy in the treatment of ragweed allergic rhinitis." A 75% one-day gain. As my boss on the trading desk used to say, "That's some sweet corn!"
Who Knew What, and When?

Several questions come to mind when looking at this deal. If the company knew that the positive data were going to be released, why did management rush to do an offering on Wednesday?

The NEJM alerts the scientific media on Fridays to what it will publish in the coming week's edition, and journal subscribers get the news on Tuesdays, rather than Wednesdays. Dynavax acknowledged it knew as early as Friday that the positive article would be published Wednesday. It also acknowledged that the stock offering deal was put together overnight.

Dynavax sold 6.2 million shares at $4.40, grossing roughly $27 million. Had it waited a day and sold at Thursday's closing price, Dynavax could have taken in nearly $48 million -- quite a difference for a company with $60 million in cash and negative operating cash flow of $34 million over the past 12 months.

Thanks Joe for your insight.

I think tomorrow will be a rebound...even though it may drop in the morning.  Time will tell.  With this kind of volume...I just can't believe it will drop into the gap.  The Street article is still playing havoc with the price...but weak hands should have dropped by now. ???

Dynavax's Universal Flu Vaccine Improves Response of Standard Flu Vaccines
Thursday October 19, 4:00 pm ET
Preclinical Studies Show TLR9 Agonists Conjugated to Conserved Viral Proteins Provide 30-Fold Dose-Sparing of Standard Vaccine Components

BERKELEY, Calif., Oct. 19 /PRNewswire-FirstCall/ -- Dynavax Technologies Corporation (Nasdaq: DVAX - News) today announced that tomorrow, Friday, October 20, at the Second International Conference on Influenza Vaccines for the World in Vienna, Austria, it is presenting new preclinical data that show its influenza (flu) vaccine can improve the immunogenicity of standard flu vaccines. Dynavax said that the data from mouse and primate models demonstrate that co-administration of Dynavax's flu vaccine with standard vaccine enhances the immune response of the standard vaccine, allows reduction of standard vaccine dosage, and provides extra layers of protection that are not strain-dependent. Dynavax's vaccine is based on the company's proprietary TLR9 agonist-based immunostimulatory sequence (ISS) technology. The work was funded in part by a $3.0 million research and development grant for a pandemic influenza vaccine from the National Institute of Allergy and Infectious Disease (NIAID), a division of the National Institutes of Health.

Implications for Future Flu Vaccine Development

"Our findings in flu give us confidence about our ability to move forward quickly with a commercial product. In Dynavax's other infectious disease vaccine programs, preclinical data accurately predicted the results of subsequent human clinical trials," said Gary Van Nest, Ph.D., vice president of preclinical research at Dynavax.

Van Nest explained, "Standard flu vaccines provide protection against viral infection by generating neutralizing antibodies against viral surface proteins (hemagglutinin, or HA and neuraminidase, or NA). When there is inadequate antibody production or a mis-match between the vaccine and circulating virus, the effectiveness of standard vaccines can be dramatically reduced. The Dynavax universal flu vaccine not only enhances the virus-blocking, or neutralizing, effects of the co-administered standard flu vaccine, but also kills virus-infected cells and prevents virus spread that can lead to pneumonia and other often fatal sequellae, in a manner that is not strain-dependent."

"Our exciting results," added Van Nest, "are produced by linking our TLR9 agonists (ISS) to the highly conserved viral proteins, nucleoprotein (NP) and the extracellular domain of matrix protein 2 (M2e). NP-ISS induces strong type-1 helper T cell (Th1) and cytotoxic T cell responses (CTL) that kill virus-infected cells. M2e-ISS induces strong cytotoxic antibody responses that also kill infected cells, limiting disease severity. In effect, even if a standard flu vaccine does not match the virus that circulates in the season, our universal flu vaccine can potentially protect against viral disease, when combined or co-administered with any standard flu vaccine."

Data from Vienna

In the presentation in Vienna, entitled, "Induction of Broadly Protective Cellular Immune Responses Using an ISS Conjugate Vaccine," Debbie Higgins, Dynavax Senior Manager, Preclinical Programs, describes the work of Dynavax and its collaborators at The Influenza Research Center, Baylor College of Medicine.

The studies suggest that by adding the ISS-linked conserved antigens to standard vaccines, it may be possible to increase the effectiveness of any standard trivalent influenza vaccine or allow dose-reduction of standard vaccines and at the same time, increase their effectiveness. The data show the enhancement of functional antibody responses against the vaccine strains, specifically:

    -- In mice, NP-ISS co-administered with standard vaccine (Fluzone®,
sanofi-aventis) provided significantly higher antibody responses with
       30-fold less Fluzone.

-- Primates (baboons) immunized with NP-ISS co-administered with standard
vaccine developed enhanced Fluzone-specific antibody responses as
measured by hemagglutination inhibition and viral neutralization
       assays.

    -- Linking ISS to a short M2e peptide (M2e-ISS) generated a strong
       M2e-specific antibody response.

In August 2006, at the "Novel Vaccines: Bridging Research, Development and Production" conference in Cambridge, MA, Dynavax presented data that showed both its flu vaccine's ability to confer cross-protective cellular immunity against widely divergent flu strains in mice and its potential as a universal flu vaccine. The data suggest that the vaccine has the potential to eliminate annual vaccinations and enable the stockpiling of vaccine for pandemic use. Specifically, the Dynavax flu vaccine was shown to protect against both "antigenic drift" and "antigenic shift." Mice immunized with the NP-ISS conjugate and then challenged with drift and shift virus strains demonstrated statistically significant lower viral titers and higher survival rates compared to mice immunized with NP alone or a PBS placebo.

Dynavax to Show Positive Flu Drug Data
Thursday October 19, 6:10 pm ET
Dynavax Technologies to Present Positive Preclinical Data on Flu Vaccine

BERKELEY, Calif. (AP) -- Drug developer Dynavax Technologies said it will present preclinical data Friday showing its flu vaccine candidate can improve the effectiveness of other flu vaccines.

The data will be presented at the Second International Conference on Influenza Vaccines for the World, in Vienna. The data from a study on involving mice and primate models showed its flu vaccine, when given with other standard vaccines, increases the immunogenicity, or the immune response, of the standard vaccines. That means it would allow for the reduction of dosages in standard vaccine and provide extra layers of protection that aren't strain-dependent.

The study was funded in part with $3 million from the National Institutes of Health as part of a grant to develop a pandemic flu vaccine. Dynavax is developing its vaccine based on a TLR9 agonist.

Shares of Dynavax fell 42 cents, or 6.4 percent, to close at $6.13 on the Nasdaq, but rebounded 37 cents, or 6 percent, to $6.50 in aftermarket activity.

A 'Pandemic' Now Declared, Focus Back on Swine Flu Stocks
On Friday June 12, 2009, 12:00 pm EDT

The H1N1 virus is officially a pandemic, and vaccine stocks are responding.

Swine flu stocks are up again today, with some extending weekly gains to more than 20%. A number of companies released good news to end the week. From small-cap players to multi-billion dollar giants, shares are up big on high volume. Novartis (NYSE: NVS) announced today that it will have a swine flu vaccine ready by this fall. Shares are up by 4% on the news.

On Thursday, the World Health Organization raised its alert level on the virus to "Phase 6," indicating that it has officially become a pandemic.
