Title: DSCO
Post by: stockpicker on June 17, 2005, 03:34:54 AM
This is a small company who is (currently) addressing a relatively
small market. The company developed precision-engineered lung
surfactant technology as Surfactant Replacement Therapies (SRT) for
respiratory diseases. Surfactants are substances that are produced
naturally in the lungs and are essential to the lungs' ability to
absorb oxygen and to maintain proper airflow through the respiratory
system. The absence or depletion of surfactants is involved in a
number of respiratory diseases.

Surfaxin, DSCO's leading product, treats neonatals who suffer from weak lungs. The company is running various clinical trials for other indications (that address
much larger markets) but I would suggest to focus ONLY on the Surfaxin.

According to the company, the neonatal mkt size is around 350,000
patients in the US which is valued at $500M-1 billion (I will mention
other markets later).

DSCO has succesfully completed 2 phase-3 trials of the Surfaxin and
received an APPROVABLE letter from the FDA. An approvable letter
means that the FDA approves the drug ONLY WHEN THE COMPANY ADDRESSES
VARIOUS ISSUES. It often happen that an approvable letter means that
a company has to do an additional clinical trial (this happened to
AMLN several years ago), however, the approvable clauses for DSCO
were regarding QC (Quality Control issues) in the manufacturing
procedures and report tracking - nothing CLINICAL.

Unfortunately for DSCO, this was (it happened in Jan 2005)the second
time that they have failed in the procedural process. Since the
company is small, the manufacturing is done by a third party and IT
IS VERY LIKELY THAT DSCO WILL FINALLY OVERCOME THIS HURDLE. They have
hired more people with experience in GMP (Good Manufacturing
Procedures) - I am sure that they can find many Teva people with this
experience.

In any case, the FDA will inspect the manufacturing facility in July
(next month) and if approved, DSCO will launch Surfaxin late this
year or early next year.

There are competitors in this not so big market which are animal-
derived surfactants (from pigs and cows). The main problems with the
animal derived products are that they are not homogenius enough (not
controlled) and that animal products increase risks associated with
transmission of infectious agents. Swine (pig) sources theorretically
can trasmit viral diseases and bovine (cow) sources can transmit BSE,
which has an incubation period of over 10 years.

Surfaxin has proven itself in clinical trials with decreased
morbidity and mortality, but has one more advantage - nebulization!
Current administration of surfactant is done by way of a liquid
bolus. The infant is first intubated with an endotracheal tube and
surfactant is poured into one lung, the patient is turned and another
aliquot is poured into the second lung. This technique is used
thousands of times a week and often it is not given a second thought,
but there are several risks involved with this process. The nebulized
form is currently in P2 studies and will likely become THE holy grail
of these treatments.

<<The Opportunity>>
Since Surfaxin showed excellent clinical results in comparison with
today's animal derived Survanta and Curosurf, it is likely that it
will capture at least 30% of the market pretty quickly which
represent annual revenue of around $200MM (in the US). It is likely
that DSCO will soon announce EU and Japan partnerships which will
possible double this revenue, so if I use a 7x revenue multiple, we
get a $3 billion mkt cap. However, I will be very conservative and
cut this number to $1 billion, which represents a pps of around $20.

DSCO is a very small company, so I think that if the big pharmas
continue their shopping of the small biotechs, it would make a lot of
sense for the company to be acquired for around $20 and the BP will
enjoy its economy of scale to maximize market share and minimize
costs.

If you look at DSCO's chart you can see the sharp drop following
January's FDA "approvable" letter. The company was running out of
money and had to do another PIPE a couple of months later at around
$6 pps. The main risk is that DSCO will fail another FDA procedural
inspection and will have to go to another PIPE (or more likely, will
be bought for cheap by a BP) - however, you got to be VERY dumb to
fail THREE times a procedural inspection.

Risk: moderate (downside: 25%)
Upside: x2 - x3 within 6 months

Technicals look nice but I am not the TA expert around this MB

good luck!

thx for the analysis stockpicker, I'll pay with a chart.  ;)  What it shows is a channel and a closed up gap.  Nothing exciting from my point of view except it is following an uptrending channel.  Volume is quiet as well.  If it is poised to make a move I would have to say that the chart is not showing it, imo.

BUTT!

With a fundamental reason - this company finally managed to submit the bureaucracy paperwork to the FDA. Remember, the FDA has approved the efficacy (and the safety) of DSCO's leading drug candidate (Surfaxin) -- so the road for the final approval is open (most likely within 5-6 months).

I am not a T/A expert, but even I can see that the chart looks bullish.

Thanks for anybody's comment

News :
Discovery Labs' Surfaxin Granted Orphan Drug Designation for the Treatment of Bronchopulmonary Dysplasia
10/28/05

WARRINGTON, Pa., Oct 28, 2005 (PRIMEZONE via COMTEX) --
Discovery Laboratories, Inc. (Nasdaq:DSCO) today announced that the Office of Orphan Products Development of the United States Food and Drug Administration (FDA) has granted orphan drug designation to Discovery's lead product, Surfaxin(r), for the treatment of Bronchopulmonary Dysplasia (also known as Chronic Lung Disease, CLD) in premature infants. Surfaxin, a precision-engineered lung surfactant replacement therapy, has received an Approvable Letter from the U.S. FDA for the prevention of Respiratory Distress Syndrome (RDS) in premature infants and is pending approval.

CLD is a costly syndrome associated with surfactant and SP-B deficiency, and the prolonged use of mechanical ventilation and oxygen supplementation, usually associated with a premature infant being treated for RDS. Presently there are no approved drugs for the treatment of CLD. These babies suffer from abnormal lung development and typically have a need for respiratory assistance -- oftentimes, for many months, as well as comprehensive care that sometimes can span many years.

The U.S. Orphan Drug Act is intended to assist and encourage companies to develop safe and effective therapies for the treatment of rare diseases and disorders. Orphan drug designation in the United States is awarded to compounds that offer potential therapeutic value in the treatment of rare diseases, defined as those affecting fewer than 200,000 Americans. If the company complies with certain FDA specifications and should the drug receive marketing approval, orphan drug designation qualifies the sponsor for seven years of marketing exclusivity, exemption from the Prescription Drug User Fee Act (PDUFA) filing fees, and tax credits related to clinical research.

Robert J. Capetola, Ph.D. President and Chief Executive Officer of Discovery commented, "Market exclusivity under this designation would mean that Surfaxin, as a precision-engineered surfactant, has the potential to become the dominant engineered surfactant for the next decade. If Surfaxin is the first product to receive marketing authorization in the United States for the treatment of Bronchopulmonary Dysplasia, orphan status would block any future similar products for this indication throughout the United States market for a prolonged period of time."

Discovery is currently conducting a Phase 2 double-blind, controlled trial (that will enroll up to 210 very low birth weight premature infants born at risk for developing CLD) to determine the safety and tolerability of administering up to 5 total doses of Surfaxin in the first 10 days of life as a therapeutic approach for the prevention and treatment of CLD. This study is designed to determine whether such treatment can decrease the proportion of infants on mechanical ventilation or oxygen or the incidence of death or CLD.

Surfaxin is a precision-engineered, peptide-containing, synthetic surfactant that is designed to closely mimic the function of natural human lung surfactant. Discovery's Surfaxin has received an Approvable Letter from the FDA for the prevention of RDS in premature infants and is pending approval. Surfactants are substances that are produced naturally in the lungs and are essential to the lungs' ability to absorb oxygen and to maintain proper airflow through the respiratory system. Premature babies are born with a lack of natural surfactant in their lungs. Without surfactant, the air sacs in the lungs collapse and are unable to absorb sufficient oxygen resulting in RDS.

Spec play. (Long Recommendation)
Stop $1.15. Speculation play. Also, CEO bought 40K shares in Nov.
Insider buying.
Discovery Labs CEO bought 40k shares at $1.01 on 11/17

From Mike Havrilla on Seeking Alpha: Last fall, Discovery Labs (DSCO) received word from the FDA that its complete response for Surfaxin was accepted, but the six-month Class II review designation means that the decision date is pushed out to mid-April. DSCO presents a model trade on a pending FDA decision at this time since it will likely enjoy a run-up in share price from current levels around $1.20 as the decision date nears. If DSCO experiences the bump in price, simply let the profits ride through the decision or just book all of the gains for a lower risk, lower reward trade on the expected increase in share price volatility heading into mid-April.

Discovery Labs Reports Fourth Quarter 2008 Financial Results
Friday , March 13, 2009 07:15ET
Discovery Laboratories

WARRINGTON, Pa., March 13, 2009 (GLOBE NEWSWIRE) -- Discovery Laboratories, Inc. (Nasdaq:DSCO) today announced financial results for the fourth quarter ended December 31, 2008.

For the quarter ended December 31, 2008, the Company reported a net loss of $8.5 million (or $0.08 per share) on 100.5 million weighted average common shares outstanding compared to a net loss of $12.0 million (or $0.14 per share) on 88.5 million weighted average common shares outstanding for the same period in 2007. Net cash burn for the fourth quarter of 2008 was $6.7 million, reflecting $7.6 million used for operating activities and $0.9 million used for debt service, offset by aggregate proceeds of $1.8 million received from the issuance of 1.4 million shares of common stock pursuant to financings under the Company's Committed Equity Financing Facilities (CEFFs).

As of December 31, 2008, the Company had cash and marketable securities of $24.8 million. Additionally, the Company has the ability (subject to certain conditions, including price and volume limitations) to raise capital under its existing CEFFs to support its business plans. As of December 31, 2008, there were approximately 15.0 million shares (not to exceed an aggregate of $25.0 million) remaining available for issuance under the December 2008 CEFF, approximately 15.6 million shares (not to exceed an aggregate of $54.9 million) under the May 2008 CEFF, and approximately 4.5 million shares under the 2006 CEFF, which will expire in May 2009. As of March 12, 2009, a maximum of $34 million is available to the Company after giving effect to applicable discounts. The Company had 101.3 million common shares outstanding as of December 31, 2008.

The decrease in net loss for the fourth quarter of 2008 as compared to the fourth quarter of 2007 is primarily due to the Company's efforts during the fourth quarter of 2008 to conserve financial resources by limiting program development activities as we await April 17, 2009, the U.S. Food and Drug Administration's (FDA) target action date to complete its review of the NDA for Surfaxin(r) (lucinactant) for the prevention of Respiratory Distress Syndrome (RDS) in premature infants. Additionally, in the fourth quarter of 2007, the Company incurred significant expenses related to the development of its Capillary Aerosolization Technology for the delivery of aerosolized KL4 Surfactant.

Robert J. Capetola, Ph.D., President and Chief Executive Officer of Discovery Labs, commented, "We believe our KL4 Surfactant and Capillary Aerosolization Technology Platform has the potential to address multiple respiratory problems in a range of patient populations from premature infants to adults. Our top priority is to gain potential FDA marketing approval for Surfaxin for the prevention of RDS in premature infants. As we await the FDA's decision, we plan to continue to conserve our financial resources while simultaneously preparing to build a significant pediatric respiratory franchise around our lead pipeline programs, Surfaxin and Aerosurf(r)."

Selected Key Priorities for 2009 include:

* We are focused on potentially gaining FDA approval for Surfaxin for
the prevention of RDS in premature infants. The FDA has established
April 17, 2009 as its target action date to complete its review. If
approved, Surfaxin will represent the first synthetic,
peptide-containing surfactant for use in pediatric medicine and the
first product from our technology platform.

* Initiate our Phase 2 clinical program for Aerosurf(r), our
proprietary KL4 Surfactant in aerosolized form, which we are
developing using our Capillary Aerosolization Technology, initially
for premature infants at risk for RDS.  Aerosurf will potentially
provide the neonatal medical community, for the first time, an
ability to treat premature infants at risk for RDS without the
risks generally associated with invasive endotracheal intubation
and mechanical ventilation.  We believe that Aerosurf holds the
promise to significantly expand the use of surfactants in pediatric
   medicine.

* Advance our ongoing Phase 2 trial of Surfaxin for children up to 2
years of age with Acute Respiratory Failure, continue our support
of a sponsor initiated Phase 2a trial at University of North
Carolina that is using our aerosolized KL4 surfactant in patients
with Cystic Fibrosis, and continue development of our novel
lyophilized KL4 Surfactant, Surfaxin LS, which is manufactured in a
dry powder form and reconstituted as a liquid just prior to
administration. Surfaxin LS, among other things, may potentially
simplify storage requirements and methods of administering
   surfactant.

* Following the potential approval of Surfaxin, we plan to establish
our own specialty pulmonary commercial organization that will focus
on creating a significant presence in the neonatal and pediatric
intensive care units. Alternatively, we would also consider a
strategic alliance or collaboration, assuming such a potential
transaction would meet our business and strategic requirements.

* Secure strategic alliances to strengthen our financial position,
support development and commercialization of our pediatric
respiratory franchise in international markets and support
development and commercialization of our adult respiratory programs
   in worldwide markets.

Readers are referred to, and encouraged to read in its entirety, the Company's Annual Report on Form 10-K for the fiscal year ended December 31, 2008, which includes full details on the Company's business plans and operations, financial condition and results of operations. Discovery Labs Advances KL4 Surfactant Technology Platform With Lyophilized Formulation
Thursday , March 26, 2009 07:00ET
Discovery Laboratories

WARRINGTON, Pa., March 26, 2009 (GLOBE NEWSWIRE) -- Discovery Laboratories, Inc. (Nasdaq:DSCO) today announces that it has created a lyophilized formulation of its synthetic, peptide-containing KL4 surfactant technology. Lyophilized KL4 surfactant is being developed to support Discovery Labs' pipeline of surfactant product candidates including Surfaxin(r) (lucinactant as a liquid formulation) and Aerosurf(r) (lucinactant as an aerosolized formulation). Discovery Labs is developing a pipeline of surfactant-based therapies targeted to address several respiratory disorders associated with surfactant deficiency or dysfunction. Lyophilized KL4 surfactant is manufactured as a dry powder formulation that is reconstituted prior to use. Key next steps include planned discussions with global regulatory agencies intended to determine the development pathway leading to the initiation of a late-stage neonatal clinical trial program.

The currently available surfactants are animal-derived liquid formulations that require constant refrigeration prior to use. Surfaxin, which is Discovery Labs' lead product that is currently under review by the United States FDA for potential approval for prevention of Respiratory Distress Syndrome (RDS) in premature infants, also requires refrigeration prior to use. Lyophilized KL4 surfactant is intended to eliminate continuous cold chain storage and refrigeration, greatly improving product flexibility and ease of use for healthcare practitioners. In addition, Discovery Labs believes that its lyophilized formulation has the potential to:

    * simplify drug administration with reduced preparation time,
    * improve drug product stability with extended shelf-life, and
    * further improve product clinical performance.

Robert J. Capetola, Ph.D., President and Chief Executive Officer of Discovery Labs, commented, "We have established that the key functional attributes of our liquid KL4 surfactant are preserved following lyophilization and are now completing development activities to support our regulatory and clinical development paths. A comprehensive assessment of development scale manufacturing lots is ongoing including full drug product characterization. These development lots are also actively being assessed under both real-time and accelerated stability protocols and are being evaluated in several proof-of-concept experiments in well established pre-clinical RDS models. Importantly, the results of this work may provide additional intellectual property to support our technology platform. We believe that our lyophilized KL4 surfactant technology could provide several technical advantages for our surfactant product candidates."

Discovery Labs is planning a worldwide, late-stage development program for Surfaxin LS(tm), the lyophilized formulation of Surfaxin, for the prevention of RDS in premature infants. To pursue regulatory approval for Surfaxin LS in the United States, Discovery Labs plans to request a pre-IND meeting with the FDA with a view to agreeing upon and potentially initiating a clinical trial with Surfaxin LS in 2010. Similarly, to gain regulatory approval for Surfaxin LS in Europe, Discovery Labs plans to request a scientific advice meeting with the EMEA with a view to agreeing upon and potentially initiating a clinical trial in 2010. If a similar clinical trial design satisfies both regulatory agencies, Discovery Labs anticipates conducting a single clinical program to gain regulatory approval for Surfaxin LS in the United States and Europe.

"We believe there is an unprecedented opportunity to improve the management of RDS throughout the world," commented Dr. Thomas F. Miller, Discovery Labs' Senior Vice President of Commercialization and Corporate Development. "The neonatal market is quite underserved today. We remain committed to the advancement of our KL4 surfactant technology platform to improve the quality of RDS management in the neonatal intensive care unit. Our three lead programs, Surfaxin, Surfaxin LS and Aerosurf, represent a completely new and comprehensive approach for RDS management in which the therapeutic solution is tailored to the unique need of a patient."
