REDWOOD CITY, Calif., June 1, 2006 /PRNewswire-FirstCall via COMTEX/ -- Threshold Pharmaceuticals, Inc. (THLD, Trade), today announced that the Independent Data Monitoring Committee (IDMC) recently evaluated the data collected to date from the Company's ongoing pivotal phase 3 clinical trial of glufosfamide for the second-line treatment of patients with metastatic pancreatic cancer. The IDMC concluded that the trial can continue to completion.

For the planned interim analysis the IDMC could have recommended stopping the trial early if there were major safety concerns in the glufosfamide arm. After reviewing the interim data the IDMC recommended to continue the study to completion.

"This is an important milestone in the development of this novel therapy," said Alan Colowick, M.D., chief medical officer at Threshold. "We anticipate announcing top-line results at the end of the fourth quarter this year. We are hopeful that, if successful in this trial, glufosfamide will give patients another treatment option against this deadly cancer."

Threshold Pharmaceuticals Up on Positive Cancer-Drug Trial Data

NEW YORK (AP) -- Shares of Threshold Pharmaceuticals Inc. leaped Thursday after the biotechnology company reported good mid-stage trial results for its drug against advanced pancreatic cancer, used with the chemotherapy drug gemcitabine.

Stock of the Redwood City, Calif.-based company jumped 91 cents, or 28.44 percent, to close at $4.11 on the Nasdaq Stock Market.

Thursday's strongest level, on heavy volume, was $4.23. On a 52-week basis, there was a high of $16.98 on April 17 and a low of $1.42 on July 24.

Threshold said that out of 28 cancer patients taking its drug, glufosfamide, 21 percent showed about a 30 percent decease in lesion size, while the progression of lesions in 36 percent of patients ceased.

For comparison, late-stage trial data for just gemcitabine-brand name Gemzar demonstrated a 5 percent response rate, according to Lazard Capital Markets analyst Joel Sendek.

Even though Threshold's glufosfamide data are encouraging, the drug nonetheless faces strong competition from OSI Pharmaceuticals Inc.'s Tarceva, and potentially from Genentech Inc.'s Avastin and Imclone Systems Inc.'s Erbitux.

"We model glufosfamide 2008-2010 U.S. sales of $11.7 million, $21.9 million, and $45.6 million, respectively," Sendek said in a research note.

Threshold doesn't have any drugs on the market. In 2005, the company posted a loss of $44.4 million, or $1.63 a share, on revenue of $690,000.

On May 12, Threshold shares dropped as much as 80 percent to $3 after the drug developer revealed the U.S. Food and Drug Administration had halted clinical trials of the company's lead drug candidate, TH-070, because of liver toxicity. That drug was for treatment of benign prostatic hyperplasia and aimed at providing rapid symptom improvement by targeting overgrown prostate cells.

The company later dropped development of the drug altogether.

On Thursday, Threshold said glufosfamide was generally well tolerated in combination with gemcitabine, but that the incidence of treatment-related toxicity to the kidneys "may be slightly higher" than in earlier observations of the drugs used individually.

Lazard Capital has had an investment-banking relationship with the company but Sendek doesn't own any shares.
