Generex Biotechnology Announces Results of a Trial Using Generex Oral-lyn(TM) in Adolescent and Young Adult Patients With Type-1 Diabetes Mellitus
Monday April 10, 9:00 am ET

Results Demonstrate Metabolic Control in Diabetes Showed a Significant Improvement When Compared to Standard Therapy; Results Presented at a Marketing Program Sponsored by Company's Joint Venture Partner, PharmaBrand SA

TORONTO--(MARKET WIRE)--Apr 10, 2006 -- Generex Biotechnology Corporation (NasdaqSC:GNBT - News), a leader in the area of buccal drug delivery and developer of Generex Oral-lyn™, the Company's proprietary oral insulin spray product, announced today interim three-month results of a long-term six-month clinical trial performed in 24 adolescents and 5 young adult patients with Type-1 diabetes mellitus (DM). This data continuously showed that replacing just 1 one daily subcutaneous injection (s.c.) of regular insulin with Generex Oral-lyn™ during the day, improves parameters of metabolic control in the same manner observed with intensively monitored standard therapy.

At the 3rd month evaluation of this six-month trial, it was demonstrated that a trend similar towards normalization exists when replacing the lunch-time dose of regular insulin with the Generex Oral-lyn™ spray. In fact, not only Daily Glucose Profiles and Fructosamine demonstrated improved values but, of special importance, Glycosylated Hemoglobin (HbA1c), the criterion standard to evaluate metabolic control in Diabetes, showed a significant improvement when compared to standard therapy. The investigators concluded, in their three-month interim analysis, that successful and efficient replacement of injected regular insulin at lunchtime was achieved with Generex Oral-lyn™ altogether with 100% compliance.

These encouraging results prompted the investigators and the partners Generex-Pharmabrand to design studies using larger number of Type-1 DM subjects and replacing the three mealtime doses of regular insulin with the Generex-Oral-lyn™ spray on subjects maintained in once daily glargine insulin therapy. These larger studies will be designed for approval in Canada and other countries.

Marketing Program sponsored by the Consortium Generex-Pharmabrand.-

The above mentioned data was presented and discussed in a seminar that included a marketing program. It was held by the Company's Joint Venture partner, PharmaBrand SA. All three-month results in safety and efficacy were discussed in Quito, Ecuador by Dr. Jaime Guevara-Aguirre M.D. during a meeting of 150 doctors (Pharmabrand sponsored this meeting as part of its post-approval marketing program). The results were received with enthusiasm by the doctors who will make use of them when Generex Oral-lyn™ is marketed and distributed nationwide in the next few weeks.

A week-long training seminar concluded Thursday, April 6, 2006, for the PharmaBrand S.A. sales force, which is preparing to begin nationwide marketing of the first commercial run of Generex Oral-lyn™ produced in Ecuador.

The April 2006 issue of Pediatrics features a study on the prevalence and correlation of depressed mood in adolescents with diabetes, entitled, "Prevalence and Correlates of Depressed Mood Among Youth with Diabetes." The results of the study showed an association between higher mean HbA1c and frequency in emergency room visits, and depressed mood. "The Pediatrics study demonstrates the vicious cycle created by a breakdown of therapy, increased complications leading to depressed moods in adolescents. Dr. Guevara-Aguirre's results shows that patient friendly Generex Oral-lyn™ leads to continuous compliance in adolescents," said Anna Gluskin, Generex's President & Chief Executive Officer.

Generex Biotechnology
Generex Biotechnology (GNBT, news, msgs) is a speculative bird-flu play, but don't let that ruffle your feathers. The company has several potential product lines, and insiders bought the stock again in March at $3.22. I'll take that as a sign the company's two main areas of research -- vaccines and a novel drug delivery platform -- hold promise.

Generex develops synthetic vaccines made from proteins that are tweaked to look like a virus. They are close enough to trick the body into thinking it has been exposed to the real deal, so they ready the immune system for an attack. But the designer proteins won't kill people -- like real vaccines can.

Another advantage is that the proteins can be made faster than normal vaccines. "This allows you to inoculate large numbers of people very quickly," says Anna Gluskin, the company's chairman and chief executive. That could come in handy if we ever need a big supply of bird-flu vaccine in a hurry. Generex is developing designer vaccines to fight HIV, skin cancer and breast cancer.

Generex also has a user-friendly drug delivery system that lets patients spray an aerosol version of medications into their mouths. The trick is that compounds in the spray block the breakdown of drug molecules long enough for the membrane inside the mouth to absorb them. The delivery platform, called "RapidMist," is already approved in Ecuador as a way to take insulin. Generex's oral insulin spray is called Oral-lyn.

Oral-lyn is still two years or so away from approval in major markets like the U.S., Canada and Europe. But Generex has $43 million in cash and a modest cash burn rate of about $1.8 million per quarter. A partnership with a bigger drug company could juice the stock before Oral-lyn hits a major market. RapidMist may also be deployed to deliver other medications, such as morphine.

Generex Biotechnology Subsidiary Receives Patent From European Patent Office
Award in Ii-Key Hybrids Technology to Regulate Antigen Presentation by Immunoregulatory Cells

TORONTO, May 01, 2006 (MARKET WIRE via COMTEX) -- Generex Biotechnology Corporation (
8:31am 05/01/2006

GNBT2.66, -0.01, -0.4%) announced today that its wholly owned subsidiary, Antigen Express, Inc., has been granted a European patent titled "Regulation of Antigen Presentation." The patent issued on December 21, 2005 relates to the use of Ii-Key peptides in regulating antigen presentation. It broadly covers the use of Ii-Key in potential immunotherapy treatments of cancer and infectious diseases.
Antigen Express is pursuing novel immunotherapeutics based on the antigen-specific stimulation of T helper cells. The use of Ii-Key to augment the potency of antigenic peptides constitutes one of two platform technologies being developed at the Company. The most advanced of these projects employs an Ii-Key modified peptide from the tumor-associated HER-2/neu protein. That compound has been in the clinic for a year in patients with breast cancer and has shown good activity in stimulating T helper cells to specifically recognize HER-2/neu. A similar vaccine employing Ii-Key and a fragment of the H5 protein from the H5N1 virus is being developed as a prophylactic vaccine for the potentially pandemic H5N1 avian influenza virus.
