ANNOUNCES POSITIVE INTERIM ANALYSIS IN PH II TRIAL:
Allos Therapeutics, Inc. (ALTH)
announced the completion of pre-planned interim analysis of patient response and safety data
from the Company's pivotal Phase II trial of PDX in patients with relapsed or refractory peripheral
T-cell lymphoma. Results of the interim analysis of patient response data exceeded the
pre-specified threshold for continuation of the trial. In addition, an independent Data Monitoring
Committee (DMC) completed an interim analysis of the safety data from the first 35 evaluable
patients and recommended that the trial continue. No major safety concerns were identified by
the DMC. The DMC will conduct another interim analysis of safety data from the first 65 evaluable
patients and the Company expects to report the outcome by the end of 2007. The Company now
expects to complete enrollment in the trial in the second quarter of 2008. The primary endpoint
of the study is objective response rate (complete and partial response). Secondary endpoints
include duration of response, progression-free survival and overall survival. The FDA granted
orphan drug designation to PDX for treatment of T-cell lymphoma in July 2006 and the Commission
of the European Communities, with a favorable opinion of the Committee for Orphan
Medicinal Products of the European Medicines Agency, granted orphan drug designation in April
2007. PDX is also being studied in a Phase II trial in patients with relapsed or refractory non-
Hodgkin's lymphoma and a Phase 1 study in patients with advanced non-small cell lung cancer.
Rituxan(R) (rituximab) is a chimeric monoclonal antibody that is currently used in the treatment
of non-Hodgkin's lymphoma. It is currently co-marketed by Biogen Idec (BIIB) and Genentech
(DNA) in the U.S. market and Roche in the EU. The Company expects to enter a Phase I trial of
another candidate, RH1, a small molecule chemotherapeutic agent, for advanced solid tumors
during the second half of 2007. Allos recently discontinued its Phase III trial of EFAPROXYNTM
(efaproxiral) plus whole brain radiation therapy in women with brain metastases originating from
breast cancer. The study failed to achieve its primary endpoint of demonstrating a statistically
significant improvement in overall survival in patients receiving EFAPROXYN plus whole brain
radiation therapy, compared to patients receiving whole brain radiation alone.
